【預(yù)期用途】

尼古丁(Nicotine)是煙草中的主要生物堿，是導(dǎo)致吸煙成癮的物質(zhì)動(dòng)因，也是評(píng)價(jià)人體攝入煙草煙霧的常用指標(biāo)。但因?yàn)槟峁哦“胨テ诙蹋瑹o法作為標(biāo)志物檢測(cè)，其代謝物可替寧因?yàn)榘胨テ陂L(zhǎng)作為吸煙和戒煙的標(biāo)志物。

本品采用競(jìng)爭(zhēng)抑制法和膠體金免疫層析技術(shù)，用于快速定性檢測(cè)人體唾液中的可替寧，適用于評(píng)價(jià)煙草煙霧攝入的初步篩查。

【主要組成成份】

• 可替寧檢測(cè)試劑盒（40人份）：每人份鋁箔袋單獨(dú)包裝。其中試劑盒由金標(biāo)可替寧單克隆抗體、可替寧-BSA結(jié)合物、羊抗兔IgG多克隆抗體、硝酸纖維素膜、聚酯纖維素膜、塑料背襯、塑料模板組成。
• 一次性滅菌唾液取樣棒（40人份）
• 唾液收集器（40人份）
• 使用說明書（1份）

• 【檢驗(yàn)方法】

• 撕開鋁箔袋，取出試劑，應(yīng)在1小時(shí)內(nèi)盡快使用。
• 拿出一次性唾液收集器。（步驟1）
• 手持收集器蓋帽使海綿頭含入口中，直至海綿頭吸滿液體（膨脹變軟）。（步驟2）
• 將海綿頭放回收集器并擰緊蓋帽，使所含樣本流入收集器下端。（步驟3）
• 將試劑盒置于干凈平坦的臺(tái)面上，同時(shí)打開滴樣孔蓋帽。（步驟4）
• 擠壓收集器上端管壁，垂直滴加2滴無空氣泡的唾液（約100ul）于加樣孔（S）中。(步驟5)
• 等待紫紅色條帶的出現(xiàn)，3-5分鐘時(shí)直接觀察結(jié)果，10分鐘后判定無效。 

2結(jié)果判定
a) 陽性反應(yīng)：局部有明顯的炎性反應(yīng)，皮厚差大于或等于0mm。
b) 疑似反應(yīng)：局部炎性反應(yīng)不明顯，皮厚差大于或等于0mm、小于4.0mm。
c) 陰性反應(yīng)：無炎性反應(yīng)。皮厚差在0mm以下。
凡判定為疑似反應(yīng)的牛只，于*次檢疫60d后進(jìn)行復(fù)檢，其結(jié)果仍為疑似反應(yīng)時(shí)，經(jīng)60d再復(fù)檢。如仍為疑似反應(yīng)，應(yīng)判為陽性。
2、其他動(dòng)物牛型結(jié)核分枝桿菌PPD皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)
參照牛的牛型結(jié)核分枝桿菌PPD皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)進(jìn)行。
二、禽型結(jié)核分枝桿菌PPD皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)
禽型結(jié)核分枝桿菌PPD皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)是zui廣泛使用于家禽的試驗(yàn)。牛及其他哺乳動(dòng)物也可用牛型結(jié)核分枝桿菌PPD和禽型結(jié)核分枝桿菌PPD同時(shí)在不同的部位進(jìn)行，以區(qū)分特異性和非特異性細(xì)菌反應(yīng)。
1、禽的禽型結(jié)核分枝桿菌PPD皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)
（1）操作方法
用10mm×0.5mm針頭肉垂皮內(nèi)注射0.1mL(0.25萬IU)禽型結(jié)核分枝桿菌PPD，48h后觀察結(jié)果。
（2）結(jié)果判定
陽性反應(yīng)為接種部位腫脹，從5.0mm直徑的小硬結(jié)到擴(kuò)展至其他肉垂與頸部的廣泛性水腫。火雞肉垂的結(jié)核分枝桿菌PPD皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)，沒有家禽的試驗(yàn)?zāi)菢涌煽俊Ｆ渌蓊愐部捎靡眭胱髟囼?yàn)，但結(jié)果一般不理想。水禽可接種腳蹼，但試驗(yàn)不敏感并經(jīng)常與接種部位的感染相混淆。
2、牛的禽型結(jié)核分枝桿菌PPD皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)
（1）操作方法
與牛型結(jié)核分枝桿菌PPD皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)相同，只是禽型結(jié)核分枝桿菌PPD的劑量為每頭0.1mL。

（2）結(jié)果判定
a) 對(duì)牛型結(jié)核分枝桿菌PPD的反應(yīng)為陽性(局部有明顯的炎性反應(yīng)，皮厚差大于或等于0 mm )，并且對(duì)牛型結(jié)核分枝桿菌PPD的反應(yīng)大于對(duì)禽型結(jié)核分枝桿菌PPD的反應(yīng)，二者皮差在2.0 mm以上，判為牛型結(jié)核分枝桿菌PPD皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)陽性。火雞肉垂的結(jié)核分枝桿菌PPD皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)，沒有家禽的試驗(yàn)?zāi)菢涌煽俊Ｆ渌蓊愐部捎靡眭胱髟囼?yàn)，但結(jié)果一般不理想。水禽可接種腳蹼，但試驗(yàn)不敏感并經(jīng)常與接種部位的感染相混淆。
