1)RT-PCR可以檢測(cè)組織、細(xì)胞、血液、細(xì)菌等很多材料,不同的樣本有不同要求,實(shí)驗(yàn)前請(qǐng)充分溝通確認(rèn)實(shí)驗(yàn)方案和樣本情況;

2)客戶(hù)盡可能提供實(shí)驗(yàn)的背景信息、物種、基因準(zhǔn)確的名稱(chēng)和ID號(hào);

3)客戶(hù)盡量不要提供DNA或RNA樣品。

4)樣本保存于液氮或干冰,也可以保存于Trizol液中。

實(shí)時(shí)熒光定量PCR(quantitative real-time PCR,qPCR)是指在PCR反應(yīng)體系中加入熒光基團(tuán),利用熒光信號(hào)積累實(shí)時(shí)監(jiān)測(cè)整個(gè)PCR進(jìn)程,*通過(guò)標(biāo)準(zhǔn)曲線對(duì)未知模板進(jìn)行定量分析的方法。實(shí)時(shí)熒光定量PCR的化學(xué)原理包括探針類(lèi)和非探針類(lèi)兩類(lèi),探針類(lèi)是利用與靶細(xì)胞序列特異性雜交的探針來(lái)指示擴(kuò)增產(chǎn)物的增加,非探針類(lèi)是利用熒光染料或者特異性設(shè)計(jì)的引物來(lái)指示擴(kuò)增的增加。運(yùn)用該技術(shù)可以對(duì)DNA、RNA樣品進(jìn)行定量(包括*定量和相對(duì)定量)和定性分析。
